I made quite a few blogposts on homebrewing. After posting on the fora, I was asked to post som new things on this subject.

micronize raw material


I will use testosterone as an example here , but any raw material can be micronized in theory.All you need to work out is what the raw hormone is freely soluble in.
1. 10 grams of testosterone powder is dissolved into either ethanol or methanol
2. allow to stand for 20 minutes
3. use a 0.22um syringe filter to remove any contaminants from the raw material
4. transfer the sterile testosterone / alcohol solution to a clean media bottle ( volume 500mls )
5. add to the 100mls of testosterone / alcohol solution 400mls of luke warm ( approximately 40c) distilled water
6.the testosterone will immediately precipitate out of the ethanol solution
7.use a coffee filter to separate the raw material from the solution
8. dry the raw material under a lamp for a period of 1 - 2 hours
9. can repeat steps 1 - 8 to obtain a 99% purity of your raw material sample

If we prefer to have large crystals of the raw material, which are easier to work with.

To accomplish this we slowly drip cool water ( approximately 5° C ) into the raw material / ethanol solution.We also try and keep the room temperature as low as possible eg task preformed in the fridge.

Therefore three factors will determine the size of how large your raw material crystals will turn out.They are :
1. room temperature
2. water temperature
3. speed at which the water hits the raw material / ethanol solution.
 

Now , to obtain a micronized version of a raw material all we have to do is reverse the above factors :

1. above normal room temperature ( > 25 c )
2. using warm water ( > 25 c )
3. poring the warm water straight into the raw material / ethanol solution

All of these factors will ensure that your final product is micronized

 Ultra-filteration is an amazing process and is very benificial. I would imagine this process being very benificial with Test Enanthate. That way you would remove the unwanted byproducts

Transdermal delivery of tadalafil. Effect of vehicles on skin permeation.

Transdermal delivery that avoids the presystemic disposition can provide an alternative to oral administration of tadalafil. Accordingly, the aim of this study was to select the best vehicle as the first step in optimization of tadalafil transdermal delivery. The vehicles were used neat or in selected binary combinations and were evaluated for drug solubilization and transdermal delivery. The drug solubility in pure vehicles were ranked as polyethylene glycol (PEG) 400 > propylene glycol (PG) > ethanol > ethyl oleate (EO) > isopropyl myristate (IPM) > water. The solubility in binary systems containing ethanol at 2:1 ratios with EO or IPM was greater than that obtained with pure ethanol, EO, or IPM. This effect could be due to the cosolvency effect. The transdermal drug delivery from pure vehicles was ranked as IPM > EO > ethanol > PG > PEG > water. The delivery from binary mixtures of ethanol with either IPM or EO was higher than that obtained from pure solvents with the delivery increasing with increasing ethanol concentration in the mixtures. The delivery from binary mixtures was synergistic rather than additive. The study thus demonstrated a potential of tadalafil transdermal delivery. Binary combinations of ethanol with either IPM or EO provided the first step forward toward the development of transdermal delivery system for tadalafil.

For those who prefer to fine-tune their figure in a doctor’s office rather than the gym, new fat dissolving injections offer a scalpel-free alternative. But are they safe?  What's injected, is illicit, and can cause bruising and weight loss? The French-imported medical technique that's all the buzz these days is called mesotherapy. Developed in 1952 in France by Dr. Michel Pistor, originally for the treatment of vascular and infectious diseases, sports injuries, and the improvement of circulation, the technique involves the injection of small amounts of various medications into the mesoderm, the layer of fat and connective tissue under the skin  . The theory is that when these small amounts of medication are injected into the mesoderm, underlying fat is melted.

Since 1952, approximately 15,000 doctors in France and South America have been using mesotherapy, and now doctors in the U.S. are rapidly jumping on the bandwagon. In August, about 40 doctors attended the first intensive course in mesotherapy offered in the U.S. Prior to this course, presented by the International Society of Mesotherapy and the Pan American Mesotherapy Society, doctors had to travel to France to be trained. Homebrewing with beta cyclodextrins

The best tool available to make nasal and sublingual steroids are derivatives of beta cyclodextrins. The one that is most readily available is hydroxypropyl-beta cyclodextrin (HPBCD). (Note: Plain beta-cyclodextrin is of little use) In case you have not heard of these, cyclodextrins are cyclic oligosaccarides (sugars) that have a hydrophilic outer surface and a hydrophobic inner surface.(3) They can be thought of as a doughnut, with the center capable of having a steroid molecule stuck inside it. The hydrophilic outer surface makes the cyclodextrin soluble in water, and when it is combined with a steroid, it can make the poorly water soluble steroid soluble as well.(3) In addition to making steroids soluble, cyclodextrins have very other important properties that make them ideal for our purpose. Cyclodextrins are known to enhance steroid delivery through biological membranes.(3) The large CDs themselves are very bad at permeating biological membranes, but they deliver the steroid to the membrane, where it partitions into the membrane, leaving the CD on the outside of the membrane.(3) The conventional penetration enhancers like alcohols or polyethylene glycol act by disrupting the lipid layers of membranes.(3) That is a big source of irritation from the old formula, and this irritation can thus be avoided by the use of CD’s. Another advantage is, once administered, the steroid is rapidly absorbed. Nearly 95% of the steroid will be absorbed within 20 minutes. Finger dosing

I had a friend (he died a few years ago) who worked in the porn industry. When he wanted to perform he took a combination of drugs. Poly-pharmacy like many bodybuilders also like to do. He took some XTC for the mindset, some “speed” for endurance, and finally a potency pill. First that was Viagra, but later when Cialis became available he took that. He also used Proviron to increase the ejaculate volume, that doesn’t only look good on the screen, but is also more satisfying. Later when he no longer worked he was still obsessed by sex and because he knew very well what worked for him and what did not, I often used him to test out some goodies.  He learned me the method of finger-dosing often used to take recreational drugs. As shown on the picture you should be careful on the amount, because a wet finger point can hold different amounts of active powder. You can use it as described  above – underneath the tongue or between the gum and cheek. A smart way of using testosterone to prevent muscle wasting

New Australian research (July 2013) suggests that a small dose of testosterone directed solely to the liver stimulates protein synthesis, likely preventing muscle loss and wasting, and potentially promoting muscle growth. The researchers believe they have developed a safe and effective treatment for men and women, that could prevent the muscle wasting associated with many chronic diseases and with ageing.

Dr Vita Birzniece and Professor Ken Ho, from Sydney’s Garvan Institute of Medical Research, showed in healthy postmenopausal women that a small dose of the male hormone testosterone prevented protein wasting. The pure crystalline testosterone, taken orally, went straight to the liver, and the dose (40mg/day) was small enough to ensure it was contained there, with no spillover to the bloodstream and other organs.

This new approach allows people to benefit from testosterone’s ability to stimulate muscle growth and increase muscle strength. At the same time, it sidesteps the side effects of testosterone when given in the usual way – administered in much larger doses by injection, gel or patch through the skin.

When testosterone is injected, it goes directly to peripheral tissues and major organs, including the brain. This can cause unwanted side effects, including aggressive behaviour and heightened libido. It can also cause heart damage, and in women induce facial hair and a deeper voice.

When taken orally in a small dose, it is absorbed through the gut and goes straight to and acts on the liver, where it is also broken down, so that no other tissue or organ is exposed.

“The novel aspect of this research is that only the liver gets tickled with testosterone. It is a new way of using an old hormone” said Professor Ken Ho.

 “This is a great advantage because it avoids the unpleasant behavioural effects of high doses of testosterone injected into the bloodstream and the masculinising effects in women.”

“We saw that low dose testosterone, taken orally, had the same magnitude of effect on whole body protein metabolism as when it is injected in larger doses in men,” added Dr Vita Birzniece.

“This is really hopeful, because if we can see the same effect on protein metabolism at this stage, we believe it will translate into the same increase in muscle mass that we see from testosterone delivered systemically, yet avoiding all the unpleasant side effects.”

The next step will be to recruit healthy men with lower testosterone levels, as well as frail people, such as patients with kidney failure.

If the researchers can see the same effect on protein metabolism in several categories of frail people, they will design a placebo controlled study, investigating the effect of oral testosterone on muscle mass and function. If the results are positive, they will recommend that this would be a novel safe treatment for muscle wasting.

“If we can reduce muscle wasting in frail elderly people, this would prevent many falls, and therefore osteoporotic fractures,” added Professor Ho.


Testosterone Replacement Therapy (TRT)

The goal of testosterone therapy is to relieve symptoms of hypogonadism and bring testosterone levels into the normal range. The research done to the diverse application possibilities helped bodybuilders and athletes to a better understanding, though some scientific studies made conclusions that made our blood boil, walking the walk or talking the talk, remember?

Testosterone Injections
Testosterone enanthate and testosterone cypionate are the two most common types of injectable testosterone. These two types of therapy are deep muscle injections that are long acting medications. The testosterone is dissolved in oil and given every one to four weeks. The newer esterification is testosterone undecanoate which dissolved in castor oil only needs to be injected once every 12 weeks. Still an injection needs to be given by a doctor or nurse and a gel can be administered by the person who needs it and is therefore preferred (cheaper and comfortable for the patient). As you can see on the graphic it also scores better stable levels.

Androgel
Androgel has become the most popular prescribed type of androgen therapy or TRT. Androgel is an alcohol-based gel that's one percent testosterone. The gel is applied to the upper arms, abdomen, or shoulders every morning and dries quickly.

If you use testosterone gel, you must be careful not to let wet gel touch the skin of anyone, especially children. Once the gel is dry, contact with other people is safe.

Two common hydrocolloids used in alcohol gel formulation are the Carbopol-series and the Polysorbate-series.  Carbopol, which is a tradename of B. F. Goodrich, is a water-soluble carboxyvinyl polymer, with a specific product example useful for alcohol gels being Carbopol 940.  Polysorbate is a polyethyleneoxide fatty acid ester, with a specific product example useful for alcohol gels being Polysorbate 80.

For any reader wishing to try at making an alcohol gel, a simple working formula is:

 Carbopol 940        0.3 %

 triethanolamine     0.4 % (needed for pH and stability control)

 ethanol            25.0 %

 water              qs

On the right a few experiments the milky white gel is a gel with powder mixed in. It’s is better to buy ultra-fine API (active pharmaceutical ingredient aka raw powder) tis is powder that is milled by a nano-mill. A nice example is Winstrol (stanozolol) available as normal base powder for tablets and a bit more expensive as ultra-fine powder meant for Winstrol-injects.

Testim
Testim uses a compound known as pentadecalactone as a base for the gel. Pentadecalactone is very good at penetrating the skin, getting more testosterone into the bloodstream. Pentadecalactone is a pheromone produced by the apocrine glands of the male armpit that serves as female attractant. Pharmaceutical preparations are not allowed to contain perfumes or scents, maybe the manufacturer added this "sexual attractant" for this reason. On the forums guys post they smear it on their clothes. Testim has that peculiar odor (pentadecalactone) some girls like, and is sticky and can feel uncomfortable on the skin.

Striant
Striant is a testosterone tablet that's not swallowed but placed between the cheek and gum. The tablet is placed on the upper gum, where it gradually softens into a waxy glob that conforms to the shape of the gum.

Striant is a popular choice for obese men, who frequently have problems with the gel penetrating the skin. Studies have reported that lower levels of DHT is associated with Striant. DHT is a testosterone by-product that contributes to prostate enlargement and hair loss.

Testosterone skin patches
Transdermal testosterone patches come in two varieties. Testoderm is applied to the scrotum, the scrotum has to be shaven and the patches don’t stick that well thus these patch is nowadays unpopular.  Androderm is applied to the upper arms, abdomen, or back.

Skin patches are an effective but not a popular form of TRT. The patches can be an inconvenience, forming rashes or blisters, irritating skin, or falling off during exercise.

 


 

New delivery systems for androgen therapy are always being formulated.  A nasal spray formulation looks promising. Not all these nasal sprays work well,  as we found out with PT141. The pharmaceutical industry tried everything to make this compound to work other then as an injectable, but without success.  Oxytocin nasal spray contains the hormone oxytocin, also known as the love or cuddle hormone. After inhaled, oxytocin has been show to induce labor contractions, increase the production of breast milk and aid in the process of bonding between mothers and their infants. Oxytocin is also released during sexual arousal and orgasm, and some companies market oxytocin nasal spray as an aid in increasing arousal. It’s a nice hormone to add to creams, gels or other formulations.

Synthetic versions of testosterone-like hormones are being developed that may have advantages over traditional TRT. Traditional medicine learns from bodybuilders and anti-aging people. These people use several different compounds at the same time. Growth Hormone is normally used together with testosterone. Others prefer Growth Hormone releasers (secratogogues) such as GHRP 6 and other peptides, just as with testosterone these means are for sale as creams in sachets or in pre-filled syringes. Peptides are new hot item produced as gels especially in Australia, where all kinds of athletes in different disciplines use them.

 

New..the nano technology  made a new transdermal system possible.

The Transdermal Delivery System (BIOLIPID B2) is a nano-emulsion that can be applied directly to the skin by a metered pump to deliver hormones into the systemic circulation. The aims of this study were to assess the efficacy of BIOLIPID B2 to deliver testosterone systemically and evaluate the short-term effects on Thyroid-Stimulating Hormone (TSH), C-reactive protein (CRP) serum levels and blood pressure in middle aged men with testosterone deficiency. An open label randomized prospective study enrolled 14 Brazilian middle aged men with testosterone deficiency. The volunteers received in the forearm 50mgof transdermal BIOLIPID B2 testosterone nanostructured formulation daily for 3 months. Confocal Raman Spectroscopy (CRS) was used to determine depthand concentration of testosterone on skin layers. Clinical parameters and serum concentrations were compared between the baseline and 3 months after treatment. Serum concentrations of testosterone, CPR and TSH changed significantly (p<0.05) after treatment. There was, also a significant change in the waist circumference, weight and body mass index (p>0.05) and no changes were observed in the blood pressure. No adverse events were attributed to this transdermal hormone replacement therapy protocol. The BIOLIPID B2  nano-emulsion formulation is safe and effective in reestablishing testosterone and Thyroid-Stimulating Hormone (TSH) serum levels.

This transdermal protocol may serve as a strategy for hormone replacement therapy in middle aged men. It should be noted that this methods is based on a new patent process. The size detection of nanoparticles by DLS is carried out in aqueous state meaning where the testosterone nanospheres are hydrated, so the diameters detected by this technique are usually larger than the non-hydrated diameters. Regarding the Zeta potential the absolute value of -42.8 mV, suggests that the nanoparticles developed in the present study are considered physically stable due to the electrostatic repulsion conferred by the chemical nature of the lipid matrix and possibly the adsorption of negatively charged ions onto the nanoparticles surface.

CONCLUSION

From the findings of this experiment we can assume that this nanostructured transdermal delivery system, have proven to be safe treating andropause symptomatology. Therefore, the present clinical data indicate that transdermal BIOLIPIDB2 testosterone (5%) could represent an interesting strategyfor Hormone Replacement Therapy. Within the limitation of this trial, it was demonstrated that the nanoemulsion is effective in reducing symptoms and was able to restore the serum levels of testosterone and TSH with significant statistical variations. These outcomes support the continued investigation of transdermal nanoparticulated hormones as a potential therapeutic agent in andropause therapy, thus. further studies are warranted to clarify its usefulness in a large sample.

Off course the new system is also tested on females: “To determine the safety and efficacy of a transdermal nanostructured formulation of progesterone (10%) combined with estriol (0.1%) + estradiol (0.25%) for relieving postmenopausal symptoms.”
